Production of pectic extracts from sugar beet pulp with antiproliferative activity on a breast cancer

摘要: In the last years, sugar beet pectins have been the subject of several investigations involving extraction methodologies, chemical composition and functional properties. The structure of pectins, which depends on the extraction method, is decisive in their capacity to induce apoptosis on several cancer cell lines like colon, prostate and breast. In this work, sugar beet pectin extraction was performed in the following steps: lipid extraction with hexane, removal of soluble complex carbohydrates and proteins, and enzymatic treatment with amyloglucosidase, protease, and pectinase. The enzymatic treatment was carried out with Rohapect DA6L under the following conditions: 50°C, pH 4.0, 2% enzyme/substrate (E/S) ratio, 15 h, and a solid to liquid ratio of 1 ∶ 10. The pectic extract showed a degree of polymerization (DP) profile of 55.8% with DP≥7; 4.9% with DP6; 5.8% between DP2 and DP6 ; 4.7% with DP2; and 28.8% with DP1. The pectic extract was examined for its antiproliferative activity on the MCF-7 breast cancer cell line. At a concentration range of 12.5–25 mg/mL the pectic extract killed 80.6% of the cells, exhibiting a higher antiproliferative activity than 4-hydroxytamoxifen (4-OHT), a classical anticancer drug, which killed 56.5% of the cells.

摘要: The antifungal effectiveness of extracts of five medicinal plant species was determined. The inhibitory activity of extracts of , , , and were evaluated against the mycelial growth of the plant pathogenic fungi , , and . All plant extracts were prepared at 60°C using solvents (either water, 50% ethanol ( ), 95% ethanol ( ), ethyl acetate or petroleum ether). Fungicidal effects of all plants tested were confirmed. Different extracts from the same plant species gave different degrees of inhibition. All aqueous extracts had weak or no activity on all fungi tested. Ethyl acetate and 95% ethanol extracts from rhizomes gave greater inhibition and a broader spectrum inhibition than the other extracts. may have potential as a new natural fungicide and may be used for the preservation of agricultural and forestry products such as fruits and vegetables.

Antibacterial properties of extracts of

摘要: Canadian goldenrod ( ) is one of the most destructive invasive weeds in South-eastern China. To evaluate its potential application as dietary supplement in red swamp crayfish ( ), the antibacterial properties of aqueous and ethanol extracts of this plant against three major pathogenic bacteria in crayfish aquaculture were examined. Inhibition zone tests and determination of minimum inhibitory concentration revealed that the extracts had lower antibacterial activity than extracts from two traditional medicinal plants that possess antibacterial properties, garlic ( ) and cortex phellodendri ( ). However, they did exhibit greater antibacterial effects than extracts from another widely used medicinal plant, , and an aquatic weed, . Aqueous extracts of Canadian goldenrod gave greater inhibition than the ethanol extracts. Crayfish fed a diet with 2% these aqueous extracts exhibited significantly higher enzyme activity of alkaline phosphatase, catalase and phenoloxidase ( <0.05). Based on the results of this study, we conclude that aqueous extracts of Canadian goldenrod are highly promising for the development of new dietary supplement for use in crayfish aquaculture.

Ethyl acetate and 95% ethanol extracts from T. chinensis rhizomes gave greater inhibition and a broader spectrum inhibition than the other extracts. T. chinensis may have potential as a new natural fungicide and may be used for the preservation of agricultural and forestry products such as fruits and vegetables.

Extracts from cotton over the whole growing season induce

摘要: Five cotton cultivars and their parents were tested for induction of germination of Wallr. (sunflower broomrape) seeds in pot and field experiments. Germination rates induced by cotton root extracts were the highest followed by stem extracts then leaf extracts. Cotton seedlings at the six-leaf stage induced higher germination than seedling at the two- and four-leaf stage, in all five cotton cultivars and their parents. In the field, the highest concentration of cotton root extracts gave the highest germination rate of , and the lowest concentration of cotton root extracts gave the lowest germination rate. Methanol extracts of cotton rhizosphere soil gave the highest germination of . In general, the root, stem and leaf extracts were more active at the six-leaf stage than other seedling stages. In conclusion, extracts of cotton rhizosphere soil and tissues have high activity in the seedling stage. Extracts of cotton across the whole growing season were able to induce germination but displayed significant cultivar interactions.

In vitro agonistic and antagonistic endocrine disrupting effects of organic extracts from waste water

摘要: Effluents from wastewater treatment plant (WWTP) have been reported to have a broad spectrum of endocrine disrupting compounds (EDCs). The majority of studies have focused on the occurrence of estrogenic activity, while ignoring nuclear hormone receptors (NRs) pathways. In the present study, a battery of in vitro yeast bioassays and a cell bioassay, including antagonistic and agonistic effects on estrogen receptor (ER), androgen receptor (AR), progesterone receptor (PR), estrogen-related receptor (ERR) and aryl hydrocarbon receptor (AHR), were conducted to evaluate the removal efficiencies of EDCs by different treatment processes of a WWTP located in Beijing. Estrogenic, anti-estrogenic, anti- androgenic, anti-progesteronic, anti-ERR and the activation of AHR activities were detected in samples from all treatment processes and the receiving water. The concentration of estrogenic contaminants with estradiol (E ) equivalent concentrations ranged from 0.82 × 10 to 3.54 × 10 g E -EQ·L . The concentration of anti-estrogenic contaminants with 4-hydroxytamoxifen (4-OHT) equivalent concentrations ranged from 1.24 × 10 to 2.36 × 10 g 4-OHT-EQ·L . The concentration of anti-androgenic contaminants ranged from 2.21 × 10 to 3.52 × 10 g flutamide-EQ·L . The concentration of anti-progesteronic contaminants ranged from 3.15 × 10 to 2.71 × 10 g RU486-EQ·L . The concentration of anti-ERR contaminants ranged from 7.09 × 10 to 6.50 × 10 g 4-OHT-EQ·L . The concentration of AHR activators ranged from 1.7 × 10 to 3.4 × 10 g TCDD-EQ·L . These processes including secondary clarifier, coagulation, as well as coal and sand filtration could eliminated 67.2% of estrogenic contaminants, 47.0% of anti-estrogenic contaminants, 98.3% of anti-androgenic contaminants, 88.4% of anti-progesteronic contaminants, 65.4% of anti-ERR contaminants and 46.9% of AHR activators. WWTP effluents contain multiple receptor disruptors may have very complex adverse effects on exposed organisms.

Evaluation of endocrine disruption and dioxin-like effects of organic extracts from sewage sludge in

摘要: Study on effective disposal and utilization of sewage sludge has recently been the target of growing interest in China. However, potential risks are associated with the use of sludge due to its contamination with toxic organics, heavy metals and pathogenic microorganisms. In this study, a screening assessment was conducted on sewage sludge samples collected from 17 different sewage treatment plants in Beijing, based on a batch of in vitro bioassays, including a set of recombinant gene yeast assays for endocrine disruption, and an ethoxy resorufin-O-deethylase (EROD) assay using H4IIE cells for aryl hydrocarbon receptor (Ah-R) agonistic activities. Our results suggested that moderate levels of estrogen receptor agonistic activities (0.9 ng E2·g to 6.8 ng E2·g , dw), but relative higher androgen receptor antagonistic activities (nd to 45%), progestin receptor antagonistic activities (nd to 80%) and Ah-R agonistic activities (1390 to 6740 pg 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD)·g , dw) were found in sewage sludge samples. However, there were no significant correlations between the toxic effects of sewage sludge and the sewage treatment processes. In addition, the 17β-estradiol (E2) equivalent quantity (EEQ) level of the sewage sludge was increased after the composting treatment, whereas the 2,3,7,8-tetrachlorodibenzo- -dioxin toxic equivalent quantity (TEQ) level of sewage sludge composted was much lower than that of sewage sludge.
